Genmab and AbbVie Report 51% Lower Lymphoma Progression Risk in Phase 3 Trial

Genmab and AbbVie reported that their Phase 3 EPCORE DLBCL-2 trial found epcoritamab, added to standard R-CHOP chemotherapy, reduced the risk of disease progression or death by 51% compared with R-CHOP alone in newly diagnosed diffuse large B-cell lymphoma patients with higher-risk scores. The improvement in progression-free survival was statistically significant, and the combination’s safety profile was generally consistent with prior experience. The investigational regimen is not yet approved for this use; the companies plan to consult global regulators and present full trial data at a future medical meeting. The results could offer a new frontline treatment option for this aggressive lymphoma.
EPCORE DLBCL-2 is the first Phase 3 trial to show a statistically significant and clinically meaningful progression-free-survival benefit from a bispecific-antibody combination in frontline DLBCL.
The global, open-label trial randomized patients 2:1: the combination arm received epcoritamab plus R-CHOP for six cycles, followed by epcoritamab alone for two cycles; the comparison arm received R-CHOP for six cycles, followed by rituximab for two cycles.
Epcoritamab is already approved in more than 65 countries for certain other lymphoma indications, marketed as Epkinly in the U.S. and Japan and Tepkinly in the EU.
DLBCL accounts for about 25–30% of non-Hodgkin lymphoma cases worldwide; roughly 25,000 new cases are diagnosed annually in the United States, according to the Leukemia and Lymphoma Society.
