Codexis Signs Evaluation Agreement to Test ECO Synthesis Platform for siRNA Production

Codexis has partnered with an unnamed siRNA drug developer to test its ECO Synthesis platform for making stereochemically defined oligonucleotide fragments through an enzymatic process. The developer will join the fragments into a final siRNA duplex, and the companies will compare the approach with conventional solid-phase synthesis on purity, product quality and assembly performance. The results will help the developer assess whether the technology could be used in future clinical development programs; Codexis says the work builds on its recent demonstration of full-length siRNA synthesis with controlled phosphorothioate stereochemistry.
Codexis CEO Alison Moore said the collaboration is an opportunity to demonstrate stereochemical control that “may offer both superior product quality and therapeutic performance” compared with assets made using traditional solid-phase chemistry.
The companies will assemble the fragments into the final duplex through ligation, which the announcement characterizes as a next-generation oligonucleotide manufacturing approach with potential benefits for scalability and efficiency as well as product quality.
Codexis said advancing ECO Synthesis is part of its strategy to industrialize scalable manufacturing solutions for complex oligonucleotide therapeutics.
